目的: 建立用高效液相-库仑阵列电化学检测器测定唾液中对乙酰氨基酚浓度的方法。 方法: 受试者服用对乙酰氨基酚片,分别于不同时间点取受试者唾液经处理,以甲醇(加5%的水相)与水相(0.03 mol·L-1 磷酸二氢钠溶液,氢氧化钠溶液和磷酸溶液调pH至6.0)(20∶ 80)为流动相,流速1 mL· min-1,350 mV电压下检测。 结果: 对乙酰氨基酚在8.3~833.3 ng·mL-1范围内线性关系良好,最低检测限为3.3 ng·mL-1,平均回收率、重复性等均符合方法学要求。 结论: 高效液相-库仑阵列电化学法操作简便、准确,灵敏度高,适用于对乙酰氨基酚体内代谢研究。

Abstract Objective: To develop an HPLC-ECD mothod for determination of paracetamol concentrations in human saliva. Method: The objects take paracetamol tablet,saliva were collected by regularly time,then concentration of paracetamol was determined by HPLC-electrochemical detector and mobile phase of methanol(5% 0.03 mol·L-1 NaH2PO4) and water(0.03 mol·L-1 NaH2PO4,pH=6.0) with a ratio of 20∶80 at flow rate of 1 mL· min-1,the detection voltage was 350 mV. Results: The content of paracetamol showed good linearity in the range of 8.3-833.3 ng·mL-1.The lowest limit of quantity was 3.3 ng·mL-1 ,the average recovery and reproducibility of related date appeared to be good. Conclusion: Electrochemical detection is convenient,accurate and sensitivity,It can be used for the study of paracetamol in vivo metabolism.
