目的: 建立土茯苓药材中落新妇苷和总黄酮的含量测定方法。 方法: 采用HPLC法测定落新妇苷的含量,使用Shim-pack VP-ODS(150 mm×4.6 mm,5 μm)色谱柱,流动相为乙腈-水-冰醋酸(18∶ 82∶ 1),流速1.0 mL·min-1,检测波长290 nm;采用比色法测定总黄酮的含量,以10%硝酸铝溶液为显色剂,测定波长为500 nm。 结果: 在选定条件下,落新妇苷测定的线性范围为0.04~2.0 μg(r=1.000),平均加样回收率为99.2%(n=9);总黄酮测定的线性范围为0.2~1.0 mg(r=0.9998),平均加样回收率为101.8%(n=9)。 结论: 本文方法简便、准确,可以用于土茯苓药材中落新妇苷和总黄酮的含量测定。

Objective: To establish methods for determination of astilbin and total flavonoids in Rhizoma Smilacis Glabrae,respectively. Methods: An HPLC method was developed for quantitative determination of astilbin in Rhizoma Smilacis Glabrae.The analysis was performed on a Shim-pack VP-ODS column(150 mm×4.6 mm,5 μm) with the mobile phase consisting of acetonitrile-water-glacial acetic acid(18∶ 82∶ 1).The flow rate was 1.0 mL·min-1 and the detection wavelength was 290 nm.A colorimetric method was used to determine the total flavanoids in Rhizoma Smilacis Glabrae.Total flavanoids were measured with 10% aluminum nitrate solution as color developing reagent and detection wavelength of 500 nm. Results: Astilbin had good linearity in the range of 0.04-2.0 μg(r=1.000),the average recovery was 99.2%(n=9).Total flavonoids had good linear relationship in the range of 0.2-1.0 mg(r=0.9998),the average recovery was 101.8%(n=9). Conclusion: The methods are simple,accurate and applicable for the quantitative determination of astilbin and total flavonoids in Rhizoma Smilacis Glabrae.
