RP-HPLC法测定牛大力药材中高丽槐素的含量

目的: 建立牛大力药材中高丽槐素的RP-HPLC含量测定方法。 方法: 采用Cosmosil 5 C₁₈-ms-Ⅱ色谱柱(4.6 mm×250 mm,5 μm),柱温30 ℃;流动相为乙腈-0.1%醋酸水溶液(43∶ 57),流速1.0 mL·min^-1;检测波长为312 nm;进样体积为20 μL。 结果: 高丽槐素进样量与峰面积呈现良好的线性关系(r=0.9999),线性范围为0.13~2.0 μg;平均加样回收率(n=6)为99.3%,RSD=1.5%。 结论: 该方法准确、快速、简便,重复性好,可用于牛大力药材的质量控制。

Objective: To establish an RP-HPLC method to determine the content of maackiain in the roots of Millettia speciosa Champ.. Methods: A Cosmosil 5C₁₈-ms-Ⅱ(4.6 mm×250 mm,5 μm) column was adopted,and the mobile phase consisted of acetonitrile and 0.1% acetic acid solution(43∶ 57) at a flow rate of 1.0 mL·min^-1.Detection was at 312 nm with injection volume of 20 μL. Results: The linear range was 0.13-2.0 μg,r=0.9999,and the average recovery(n=6) was 99.3% with RSD of 1.5%. Conclusion: This method is accurate,quick,simple and reproducible,which can be used for the quality control of the roots of Millettia speciosa.
