目的: 建立钙黄绿素脂质体包封率的测定方法。 方法: 采用阴离子交换树脂微型柱分离脂质体与钙黄绿素;紫外可见分光光度法测定钙黄绿素的含量,检测波长为484 nm,计算包封率。 结果: 钙黄绿素浓度在1~10 mg·L-1范围内线性关系良好(r=0.9999);阴离子交换树脂能够吸附浓度在0.50~5.00 g·L-1之间的钙黄绿素,柱通过率为0%;脂质体能够通过阴离子交换树脂柱而不被吸附,柱通过率在94.5%以上。利用阴离子交换树脂柱可以使脂质体与钙黄绿素得到良好的分离。利用该方法测得的钙黄绿素脂质体包封率为29.3%,RSD为1.6%(n=6)。 结论: 阴离子交换树脂-微柱离心法可以作为钙黄绿素脂质体包封率的测定方法,该方法简便快速,结果重复性好。

Objective: To develop a method for determining the entrapment efficiency of calcein liposome. Methods: Anion exchange resin was applied to separate the free calcein from the liposomes;The assay of calcein was carried out using ultraviolet-visble spectrophotometry and the entrapment efficiency was calculated.The detective wavelength was set at 484 nm. Results: The UV-VIS method had good linearity in the range of 1-10 mg·L-1(r=0.9999).The free calcein(0.50-5.00 g·L-1) was well adsorbed by the anion exchange resin and the column passing rate was 0%.The liposomes could through the minicolumn and the column passing rate was beyond 94.5%.The free calcein could be well separated from the liposomes by the anion exchange resin column.The entrapment efficiency of calcein for liposomes was 29.3% with RSD of 1.6%(n=6). Conclusion: This method is technically simple,rapid and low cost to be used for the determination of entrapment efficiency of calcein liposomes.
