目的: 观察姜黄素对大鼠局灶脑缺血/再灌注后神经细胞凋亡的作用及其机制。 方法: 采用SD大鼠线栓模型,动物随机分为3组:假手术组(sham组)、缺血/再灌注组(vehicle组)、姜黄素100 mg·kg-1组(Cur 100组),TUNEL法检测皮层神经细胞凋亡;Western-blot检测凋亡诱导因子(AIF)和细胞色素C(Cytc)在缺血皮层的表达;同时检测膜电位的变化。 结果: I/R组凋亡细胞多于sham组及Cur 100组,Cur 100能够显著改善I/R所引起的线粒体膜电位下降;降低AIF,Cytc的表达。 结论: 姜黄素能够显著减少缺血性神经细胞凋亡的发生,可能与保护线粒体功能,减少AIF和Cytc的释放有关。

Objective: To investigate the effect of curcumin on middle cerebral artery occlusion rats and its mechanism. Method: Cerebral ischemia reperfusion(IR)injury were used induce MCAO in rats.Administration of curcumin 100 mg·kg-1,i.p.30 min.after MCAO,sham group and vehicle group were treated with equal vehicle respectively.24 h after MCAO,cortex was harvested to analyze apoptosis with Tunnel staining.Also,Western-blot was used to study the expression of AIF and Cytc proteins.Meanwhile,mitochondrial membrane potential was assayed. Results: Curcumin significantly attenuated apoptosis induced by MCAO.AIF and Cytc protein expression were reduced by curcumin.Also,the increased mitochondrial membrane potential was restored by curcumin. Conclusion: Curcumin can significantly attenuated apoptosis through reduction of AIF and Cytc released from mitochondria.
