目的: 建立HPLC法测定人血浆中依托咪酯的浓度。 方法: 采用Hypersil ODS C18色谱柱(250 mm×4.6 mm,10 μm),以甲醇-0.062%醋酸胺溶液(60∶ 40)为流动相,流速为1.0 mL·min-1,柱温为室温,检测波长240 nm。 结果: 依托咪酯在0.025~2.5 μg·mL-1内呈良好的线性关系(r=0.9998),依托咪酯最低检测浓度为12 ng·mL-1,方法回收率在96.8%~102.5%,萃取回收率在80.7%~85.0%。日内、日间RSD(n=3)均小于5.9%。 结论: 本方法准确、可靠,可用于人体血浆中依托咪酯的浓度测定。

Objective: To develop an HPLC method for the determination of etomidate in human plasma. Method: The Hypersil ODS C18 (250 mm×4.6 mm,10 μm) was used as analytical column with a mobile phase consisted of methanol-0.062%ammonium acetate solution(60∶ 40).The flow rate was 1.0 mL·min-1and detection wavelength was set at 240 nm. Results: The linear ranges were 0.025-2.5 μg·mL-1 for etomidate in human plasma.The correlation coefficients were 0.9998.The etomidate low limit of detection was 12 ng·mL-1.The method recoveries were 96.8%-102.5%.The extraction recoveries were 80.7%-85.0%.Within-day and between–day RSD(n=3)were less than 5.9%. Conclusion: The method is reliable and sensitive for determination of etomidate in human plasma.
