小柴胡颗粒中柴胡皂苷含量评价方法研究

目的: 建立小柴胡颗粒中柴胡皂苷的含量评价方法。 方法: 采用薄层色谱扫描法,以三氯甲烷-甲醇-水(30∶ 10∶ 1)为展开剂,2%对二甲氨基苯甲醛40%硫酸溶液为显色剂,扫描波长λ=538 nm,光束狭缝6.0 mm×0.45 mm,同时测定柴胡皂苷a(d)、b2(b1)的含量,并统计四者之和。 结果: 在薄层色谱中阴性样品无干扰,柴胡皂苷a(d)、柴胡皂苷 b2(b1)的线性范围分别为1.30~6.50 μg(r=0.996)和1.32~6.61 μg(r=0.999);平均回收率(n=6)分别为100.2%(RSD=2.2%)和99.58% (RSD=2.0%)。实测市售商品12批,柴胡皂苷a (d)、柴胡皂苷b2(b1)的含量分别为0~2.9和0~2.6 mg·袋-1,四者含量之和为0~5.5 mg·袋-1。 结论: 本方法简单、准确,专属性强,重复性好,可作为小柴胡颗粒质量评价的指标之一。

Objective: To develop a method for evaluation of saikosaponins determination in Xiaochaihu granules. Methods: The determination was simultaneously performed by the thin layer chromatography scanning method (TLCS).The developer was CHCl3-CH3OH-H2O(30∶ 10∶ 1),40% sulphuric acid with 2% paradime thylaminobenzaldehyde was used as colour developing agent.The detection wavelength was 538 nm and narrow slot was 6.0 mm×0.45 mm,sum the contents of saikosaponin a(d) and b2(b1). Results: The linear ranges of saikosaponin a(d) and saikosaponin b2(b1) were 1.30-6.50 μg (r=0.996) and 1.32-6.61 μg (r=0.999) respectively;the average recovery rate (n=6) was 100.2% (RSD=2.2%) and 99.58% (RSD=2.0%) respectively.12 batch of samples were determined,the contents of saikosaponin a(d) and saikosaponin b2(b1) were 0-2.9 mg and 0-2.6 mg in each bag of Xiaochaihu granules.The total contents of the four saikosaponins range from 0 mg to 5.5 mg per bag. Conclusion: The method is simple,precise,specific and reliable,can be used as one of quality index for Xiaochaihu granules.
