HPLC法测定华山参滴丸中消旋山莨菪碱、_东莨菪碱及阿托品的含量___

目的: 建立HPLC法同时测定华山参滴丸中消旋山莨菪碱、东莨菪碱及阿托品的含量。 方法: 采用资生堂Capcell-Pak C₁₈(4.6 mm×250 mm,5 μm)色谱柱,以乙腈-醋酸盐缓冲液 (5∶ 95)为流动相A,乙腈-醋酸盐缓冲液 (15∶ 85)为流动相B,进行梯度洗脱(0~30 min,60% A→35% A;30~40 min,35% A→30% A;40~44 min,30% A→20% A;44~45 min,20% A→60% A;45~60 min,60% A),流速1.0 mL·min^-1,检测波长210 nm;柱温30 ℃。 结果: 消旋山莨菪碱、东莨菪碱及阿托品的线性范围分别为0.08~1.82,0.07~1.65,0.04~1.03 μg(r=0.9999);平均回收率分别为99.1%(RSD=1.8%),97.0%(RSD=1.6%),104.0%(RSD=0.84%)。 结论: 所建立的HPLC法可同时测定华山参滴丸中消旋山莨菪碱、东莨菪碱及阿托品的含量。

Objective: To develop an HPLC method for determination of raceanisodamine,scopolamine and atropine in Huashanshen dripping pills. Methods: The analysis was carried out on a Capcell-Pak C₁₈(4.6 mm×250 mm,5 μm)column;The mobile phase was composed of acetonitrile-acetate buffer solution(5∶ 95)(A)and acetonitrile-acetate buffer solution(15∶ 85)(B)with gradient elution(0-30 min,60% A→35% A;30-40 min,35% A→30% A;40-44 min,30% A→20% A;44-45 min,20% A→60% A;45-60 min,60% A)and the flow rate was 1.0 mL·min^-1;The detection wavelength was 210 nm ,and the column temperature was 30 ℃. Results: The linear ranges of raceanisodamine,scopolamine and atropine were 0.08-1.82,0.07-1.65,0.04-1.03 μg(r=0.9999),respectively;The average recoveries(n=6)were 99.1%(RSD=1.8%),97.0%(RSD=1.6%),104.0%(RSD=0.84%),respectively. Conclusion: This method is simple and can be used to determine the above three components with satisfactory accuracy and repeatability.
