薄层扫描法同时测定人工牛黄中胆酸及猪去氧胆酸的含量___

 

 

目的：同时测定人工牛黄中胆酸及猪去氧胆酸的含量。方法：以甲醇为提取溶剂，制备供试品溶液；应用薄层色谱法，使用铝基硅胶G高效薄层板，以环己烷-乙醚-冰醋酸(10∶5∶5)为展开剂，10%磷钼酸乙醇溶液为显色剂；采用单波长吸收扫描法，λ=650 nm，光束狭缝10.0 mm×0.2 mm。结果：胆酸，猪去氧胆酸的线性范围均在0.2~0.6 μg之间(r≥0.99)；平均回收率(n=6)分别为101.3%(RSD=2.6%)和101.6%(RSD=2.5%)。结论：该方法专属性强，重复性好，适用于人工牛黄中胆酸及猪去氧胆酸的含量测定与质量控制。

 

Objectives:To determine simultaneously free cholic acid and hyodeoxycholic acid in artificial Calculus Bovis.Methods:Methanol was used as extraction solvent.Silica gel prefabricate thin layer plate was used with cyclohexane–ethylether-glacial acetic acid (10∶5∶5) as the developing solvent and 10% ethanol solution of phosphomolybdic acid as color reagent.TLCs single wave-length absorption was used,λ=650 nm,narrow slot:10.0 mm×0.2 mm.Results:The linear ranges of cholic acid and hyodeoxycholic acid were 0.2-0.6 μg(r≥0.99);The average recoveries(n=6) was 101.3%(RSD=2.6%)和101.6%(RSD=2.5%),respectively.Conclusions:This method has good specificity,reproducibility and is suitable for the determination and quality control of cholic acid and hyodeoxycholic acid in artificial Calculus Bovis.
