小儿咽扁颗粒的定性定量方法研究___

目的:研究建立小儿咽扁颗粒的定性定量方法。方法:采用TLC法对金银花、射干、麦冬、金果榄、玄参分别进行定性鉴别;采用HPLC法对金银花中绿原酸进行定量分析,色谱柱为Diamonsil C18 (4.6 mm×250 mm,5 μm)柱,流动相为乙腈-0.4%磷酸溶液(10∶90),检测波长327 nm,流速1.0 mL•min-1,柱温30 ℃。结果:薄层色谱斑点清晰,专属性强;绿原酸进样量在0.0198~0.992 μg范围内呈良好的线性关系(r=1.000),平均回收率为100.7%(RSD=0.7%,n=6)。结论:所建立的方法简便、准确,重复性好,可作为小儿咽扁颗粒的质量控制方法。

Objective: To study and establish the quality and quantity methods of Xiao’er Yanbian granules. Methods: Flos Lonicerae Japonicae,Rhizoma Belamcandae,Radix Ophiopogonis,Radix Tinosporae,and Radix Scrophulariae(代改请核) were identified by TLC,and the contents of chlorogenic acid in Flos Lonicerae Japonicae were determined simultaneously by HPLC. The chromatographic column was Diamonsil C18 column (4.6 mm×250 mm,5 μm) ;the mobile phase was acetonitrile-0.4% phosphoric solution (10:90) at the flow rate of 1.0mL•min-1, and the detecting wavelength was 327 nm, column temperature was 30 ℃.Results:The TLC identification was highly specific and spots were clear. Chlorogenic acid showed a good linear relationship at a range of 0.0198-0.992 μg(r=1.000),and the average recovery was 100.7% with RSD of 0.7%(n=6).Conclusion:The method established is simple,accurate and reproducible, and can be used for the quality control of Xiao’er Yanbian granules.
