RP-HPLC法同时测定红花中羟基红花黄色素A与红花黄色素A的含量___

RP-HPLC determination of safflomin A and safflower yellow A in Carthamus tinctorius L.

目的:建立红花中羟基红花黄色素A与红花黄色素A的含量测定方法。方法:采用Akasil C18(4.6 mm×250 mm,5 μm)色谱柱,以乙腈(A)-0.4%磷酸(B)为流动相,梯度洗脱(0~30 min,10%A 30~50 min,15%A 50~55 min,25%A 55~60 min,10%A),流速0.7 mL•min-1,检测波长403 nm,柱温30 ℃。结果:羟基红花黄色素A、红花黄色素A进样量的线性范围分别为0.451~1.05 μg(r=0.9993)和0.278~0.650 μg(r=0.9996)(n=5) 平均加样回收率(n=6)分别为100.6%和103.0%。结论:本方法快速、灵敏,重复性好,适用于红花中羟基红花黄色素A与红花黄色素A的含量测定。

Objective:To establish an RP-HPLC method for simultaneous determination of safflomin A and safflower yellow A in Carthamus tinctorius.Methods:The analytical column was Akasil C18 column(4.6 mm×150 mm,5 μm).The mobile phase was composed of acetonitrile(A) and 0.4% phosphoric acid(B) with gradient elution(0-30 min,10%A 30-50 min,15%A 50-55 min,25%A 55-60 min,10%A) at a flow rate of 0.7 mL•min-1.The wavelength of UV detector was 403 nm and column temperature was 30 ℃.Results:The linear ranges of safflomin A and safflower yellow A were 0.451-1.05 μg(r=0.9993) and 0.278-0.650 μg(r=0.9996) (n=5),respectively Their average recoveries(n=6) were 100.6% and 103.0%,respectively.Conclusion:This method is sensitive,repeatable and suitable to determine the contents of safflomin A and safflower yellow A in Carthamus tinctorius.
