四川盆地麦冬种质资源RP-HPLC特征图谱的建立___

目的:建立四川盆地麦冬种质资源的HPLC特征图谱。方法:采用高效液相色谱法,以槲皮素作为内标参照物,甲醇-乙腈-0.2%醋酸水溶液为流动相梯度洗脱,流速1.0 mL•min-1,检测波长280 nm,柱温30 ℃。结果:24批四川盆地麦冬资源共有36个特征峰,32~36号峰分别被鉴定为甲基麦冬黄烷酮A、甲基麦冬黄烷酮B、大黄酚、麦冬黄烷酮A、麦冬黄烷酮B。结论:所建立的四川盆地麦冬种质资源RP-HPLC特征图谱特征性及专属性强,具有可靠性 不同产区麦冬资源的化学成分差异较大,可为其优质资源筛选及质量控制研究提供参考。

Objective:To establish chromatographic specific chromatograms of germplasm resource of Ophiopogon japonicus in Sichuan basin.Method:HPLC was applied on a C18 column with mathanol-acetonitrile -0.2% acetic acid solution by gradient elution at the flow rate of 1.0 mL•min-1,and the UV absorbance was monitored at 280 nm,the temperature of C18 column was 30 ℃.Quercetin was used as the internal standard.Result:36 characteristic peaks in the 24 batches of Ophiopogon japonicus of Sichuan basin were picked up,The numbers of 32-36 were methylophiopogonanone A(67.49 min),methylophiopogonanone B(68.36 min),chrysophanol(70.63 min),ophiopogonanone A(73.95 min),ophiopogonanone B(74.51 min).Conclusion:The HPLC specific chromatograms by this method is reliable,feasible in analysis of Ophiopogon japonicus in Sichuan basin To the samples from different habitats,their content of chemistry composition are great differently,and this method can be able to provide a reference to screening for good quality Ophiopogon japonicus germplasm resources and control the quality of Ophiopogon japonicus.
