多波长高效液相色谱法同时测定桂枝汤中5种有效成分的含量___

目的:利用多波长高效液相色谱法,建立同时测定桂枝汤中5种有效成分(芍药苷、甘草苷、肉桂酸、桂皮醛和甘草酸)含量的检测方法。方法:采用Diamond C18色谱柱(4.6 mm×250 mm,5 μm),柱温25 ℃;0.1%磷酸溶液(A)-乙腈(B)为流动相,以1 mL·min-1梯度洗脱;检测波长:230 nm(检测芍药苷)、250 nm(检测甘草酸)和280 nm(检测肉桂酸、桂皮醛及甘草苷)。结果:在60 min内桂枝汤中芍药苷、甘草苷、肉桂酸、桂皮醛和甘草酸5种有效成分被完全分离;峰面积与其浓度呈良好的线性;加样回收率(n=5)分别为98.7%(RSD=1.4%),101.7%(RSD=2.1%),97.1%(RSD=2.4%),96.4%(RSD=2.5%),99.4%(RSD=1.9%)。结论:利用多波长高效液相色谱法,建立了一种可靠的同时测定桂枝汤中5种有效成分含量的检测方法。

Objective: To develop an HPLC method with multiple wavelength ultraviolet detection for the simultaneous determination of peoniflorin,liquiritin,cinnamic acid,cinnamal and glycyrrhizic acid in Guizhi decoction.Methods:The samples were separated on a Diamond C18 column(4.6 mm×250 mm,5 μm),by a gradient elution using 0.1% phosphoric acid(A) and acetonitrile(B) as mobile phase at a flow rate of 1.0 mL·min-1.The eluate was detected by multiple wavelength:230 nm for peoniflorin;280 nm for liquiritin,cinnamic acid and cinnamal and 250 nm for glycyrrhizic acid.Results:The complete separation was obtained within 60 min for the five compounds(peoniflorin,liquiritin,cinnamic acid,cinnamal and glycyrrhizic acid).Five regression equations showing linear relationships between peak-area and content of each compound were obtained.The average recoveries(n=5) of the compounds listed above were 98.7%(RSD=1.4%),101.7%(RSD=2.1%),97.1%(RSD=2.4%),96.4%(RSD=2.5%),99.4%(RSD=1.9%),respectively.Conclusion:Very satisfactory and reproducible results are obtained for the simultaneous determination of the five compounds in Guizhi decoction.
