Objective:To determine the contents of Pinellia ternate in different growing areas.Methods: Amino acids in the injections were pretreated with AccQ-FlourTM separately and their derivatives were analyzed on Waters 1525 HPLC using AccQ-Tag column. Mobile phase consisted of AccQ·Tag concentrated liquid-water(110)(A) and acetonitrile-water(6040)(B) with gradient elution.The detection wavelength was 248 nm,column temperature 37 , and injection volume was 10 μL.Results:The linear regression equation was greater than 0.9997 in 17 amino acids, the average recovery(n=6) was 98.7% with RSD of 1.8%.17 amino acids from different growing areas were detected, the total content of amino acids in Hubei (19.18%) was the highest and was significantly higher than that of Henan and Guizhou two origin(P<0.05),but the total content of amino acids in Hubei was no significant difference that it compared with that of Sichuan growing areas(P>0.05).Conclusion:This method is rapid,simple and accurate. The total content of amino acids of Pinellia ternate in different growing areas is significant difference.
