Objective:To separate and purify of chlorogenic acid from the crude extract of Flos Lonicerae by high-speed counter-current chromatography(HSCCC).Methods:Chlorogenic acid was extracted with the assistance of microwave power,then filtrated and concentrated under reduced pressure.The condensed sample was separated by HSCCC.Two-phase solvent system containing nbutanol-acetic acid-water(4 ∶1∶5)was run on a preparative scale.The lower phase was used as the mobile phase and the upper was used as the stationary phase in the head to tail elution mode.The flow rate was 2.0 mL·min-1.The running rate was 1000 r·min-1 at 25 ℃.Results:Chlorogenic acid with 98.2% purity was obtained by once separation.Conclusion:The HSCCC method is simple,high-performance and suitable for isolating and purificating chlorogenic acid from Flos Lonicerae.
