大鼠尿中拉米夫定的毛细管电泳分析方法的建立和应用___

Objective:To develop and validate a capillary electrophoretic method for the determination of lamivudine excretion in rat urine.Method:Rat urine samples were deproteinized with acetonitrile and separated on an untreated fused silica capillary(45 cm×75 μm,effective length is 36.7 cm)with a buffer consisting of 20 mmol·L^-1 phosphate buffer(pH 2.5).The applied voltage was 15 kV.The UV detection wavelength was set at 270 nm and the capillary was kept at 25 ℃.Results:The calibration curve for lamivudine was linear over the concentration range of 20.0-1000 μg·mL^-1(r=0.9994).The intra- and inter-day precisions were less than 5.8% and the accuracies were 93.8%-101.9%.The extraction recoveries of lamivudine were 90.3%-94.8%.The urine cumulative excretion percentage of lamivudine within 24 h after i.g. administration of lamivudine to rats was 49.8%.Conclusion:The method is simple,rapid and economic,and suitable for the determination of lamivudine excretion in rat urine.
