HPLC法测定不同市售荷叶药材中金丝桃苷和异槲皮苷的含量___

Objective:To establish an HPLC method for the determination of hyperin and isoquercitrin in Nelumbo nucifera.Methods:The HPLC determination was carried out by using a Nova-Pak C₁₈ column(3.9 mm×150 mm,5 μm) and mixture of acetonitrile-0.1% methanoic acid water solution(13∶87) as mobile phase at the flow rate of 1.0 mL·min^-1.The detective wavelength was set at 255 nm and the column temperature was 34 ℃.Results:The assay displayed good lineary,the linear range of hyperin was 0.056-0.28 μg and that of isoquercitrin was 0.037-0.26 μg.The average recoveries were 96.1%(RSD=2.7%) and 98.8% (RSD=2.6%) for hyperin and isoquercitrin,respectively.The contents of hyperin and isoquercitrin in 46 Nelumbo nucifera samples of different sources were 0.35%-1.47% and 0.10%-0.69%.Conclusion:The method is simple,accurate and can be used to determine hyperin and isoquercitrin in Nelumbo nucifera.The contents of the two components in 46 market samples varied notably,and should be taken into the quality control of Nelumbo nucifera.
