摘要 目的:建立RP–HPLC法同时测定五味子油中五味子甲素与五味子乙素的含量。方法:采用kromasil C18柱(4.6 mm×250 mm,5μm)进行分离,流动相为甲醇-乙腈-水 (体积比为43∶32∶25),流速1.0 mL•min-1,检测波长254 nm,柱温30℃。结果:五味子甲素和五味子乙素浓度分别在1.83~27.45mg•L-1(r = 0.9997,n = 7),4.52~33.9mg•L-1(r = 0.9995,n = 7)范围内与峰面积呈良好线性关系;平均回收率分别为102.2%、99.3%,RSD均小于3%。结论:本法准确、快速、灵敏度高,可用于五味子油质量控制。

Abstract Objective : To develop a RP-HPLC method for the determination of deoxyschizandrin and γ-schizandrin in Schisandra chinensis(Turcz.)Baill.oil. in different areas. Methods: A kromasil C18 column(4.6mm×250mm, 5μm) with the mobile phase consisted of methonal-acetonitrile-water(V∶V∶V=43∶32∶25) was used. The flow rate was 1.0 mL•min-1, the UV detection wavelength was set at 254 nm and the column temperature was 30 ℃. Results: The calibration curve of deoxyschizandrin and γ-schizandrin was linear in the ranges of 1.83~27.45mg•L-1(r= 0.9997,n=7) and 4.52~33.9mg•L-1(r=0.9995,n=7)respectively. The average recoveries of deoxyschizandrin and γ-schizandrin were 102.2% and 99.3% respectively with RSD less than 3%. Conclusions : The method is sensitive, accurate, rapid and is suitable for the quality control of Schisandra chinensis(Turcz.)Baill.oil.
