活性艳红比色法测定壳聚糖含量___

目的:建立一种简单快捷地检测壳聚糖含量的方法。方法:利用活性艳红3B-A与壳聚糖反应生成的络合物的光谱特征,选择在波长575 nm处测定溶液中壳聚糖含量。结果:在pH 3.2的甘氨酸缓冲体系中,壳聚糖/活性艳红体积比为5∶1的条件下,壳聚糖含量在0~80 mg•L-1范围内与吸光度呈现良好的线性关系(r=0.9995),平均回收率为98.0% (RSD为0.57%,n=7)。结论:该方法快速、简单且灵敏度高,适用于测定溶液中壳聚糖的含量。

Objective:To develop a simple and fast spectrophotometric method for determination of chitosan.Method:On the basis of the spectrometry feature for the Cibacron Brilliant Red-Chitosan complex,the absorbance at 575 nm was in proportion to chitosan concentration and was chosen as its determination wavelength.Results:Under the condition of pH 3.2 glycine buffer solution and chitosan/Cibacron Brilliant Red 3B-A volume ratio of 5∶1,the concentration of chitosan was linear (r=0.9995) with in 0-80 mg•L-1.The average recovery ratio was 98.0% (RSD was 0.57%,n=7).Conclusion:This proposed method is fast,simple and of good sensitivity.It can be used for the determination of chitosan contents in solution.
