Objective:To establish a GC/MS method for the determination of nicotine in tobacco.Methods:Nicotine in tobacco was extracted with ethyl acetate and analyzed on a DB-5MS column(30 m×0.32 mm×0.25 μm) with programming temperature from 60 to 280 ℃. Helium was used as carrier gas. Selected ion monitoring mode was used with the target ion of m/z 84. Quantitative analysis was performed by external standard method. Results:The calibration curve was linear in the range of 0.01-10 mg·mL-1(r=0.9998,n=6);The average recovery was 98.3%(RSD=2.0%,n=5). The limit of detection was 10 pg. Conclusion:This method is sensitive, accurate, repeatable,easy to operate and can be used for the nicotine content of tobacco.
