Objective:To develop an HPLC method for simultaneous determination of euphol and tirucallol in Radix Kansui.Methods:The sample was extracted with ethyl acetate by ultrasonication. HPLC analysis was performed on an Agilent XDB C8 column(150 mm×4.6 mm,5 μm).The mobile phase was the system of acetonitrile-water(95∶5). The flow rate was 1.0 mL·min-1 and UV detection was set at 210 nm.Results:Euphol and tirucallol had good linearity in the ranges of 0.43-6.34 μg(r=0.9999),0.21-3.17 μg(r=0.9999),respectively;The average recoveries(n=9) were 100.0% and 102.2%,respectively.The contents of euphol and tirucallol in Radix Kansui were 0.18%-0.68% and 0.12%-0.39%,respectively.Conclusion:The developed method is accurate with high repeatability,which is helpful for the determination of euphol and tirucallol in Radix Kansui.
