Objective:To establish an HPLC method for determination of ergosta-4,6,8(14),22-tetraen-3-one in Polyporus umbellatus.Methods:P. umbellatus was extracted with methanol by ultrasound-assisted extraction and the chromatographic procedure was performed with an Inertsil ODS-3(250 mm×4.6 mm,5 μm) column,the mobile phase was methanol-water(96∶4) at a flow of 1.0 mL·min-1,and the UV detection was set at 350 nm.The temperature of column was 30 ℃.Results:The linear range of ergosta-4,6,8(14),22-tetraen-3-one was 2.5-35 μg·mL-1(r=0.9997),and the average recovery(n=5) was 100.5% and RSD was 2.5%.Conclusion:The developed method is simple,accurate and reproducible,which can offer references for the quality control of P. umbellatus.
