配伍煎煮对加味二妙方中盐酸小檗碱转移率的影响___

目的:探讨加味二妙方配伍煎煮对关黄柏中盐酸小檗碱转移率的影响。方法:以盐酸小檗碱含量为指标,考察加味二妙方中其他药味与关黄柏配伍煎煮时盐酸小檗碱的转移率变化,并通过酸化或碱化考察药液的pH对盐酸小檗碱转移率的影响。结果:关黄柏与知母、苍术和蒲黄等分别配伍煎煮,药液的pH与盐酸小檗碱的转移率呈负线性相关;而全方煎煮时,药液中盐酸小檗碱的含量较低,加入适量的酸碱均可以提高转移率。结论:关黄柏与其他药味进行单独配伍煎煮时,盐酸小檗碱的转移率大小主要受pH的影响;而在全方煎煮时,受形成复合物沉淀的影响,盐酸小檗碱含量下降显著。

Objective: To explore the effects of decoction on the transferring rate(TR) of berberine hydrochloride from Cortex Phellodendri Amurensis in Jiawei Ermiaofang(i.e. double wonder TCM prescription). Methods:The content of berberine hydrochloride in the decoction solution was used as the indicator to investigate the TR of berberine hydrochloride from Cortex Phellodendri Amurensis in different decoction combination of the ingredients in Jiawei Ermiaofang with Cortex Phellodendri Amurensis. The influence of decoction solution pH on the TR of berberine hydrochloride was further investigated by adding inorganic acidic or basic regulators. Results:In decocting Cortex Phellodendri Amurensis with Rhizoma Anemarrhenae,Rhizoma Atractylodis and Pollen Typhae respectively,the lower the pH of the decoction solution shows the higher the TR of berberine hydrochloride from Cortex Phellodendri Amurensis. Adding acidic or basic regulators can both raise the TR of berberine hydrochloride in Jiawei Ermiaofang decoction. Conclusion:The TR of berberine hydrochloride from Cortex Phellodendri Amurensis is significantly influenced by the pH on decocting with other ingredients. Precipitation in complex form is the important reason for the especially low content of berberine hydrochloride in Jiawei Ermiaofang decoction.
