狭叶柴胡地上部分HPLC特征色谱研究及HPLC-ESI-TOFMS定性分析___

目的:建立不同产地狭叶柴胡地上部分黄酮类成分高效液相指纹图谱。方法:采用Kromasil C18 (4.6 mm×250 mm ,5 µm) 色谱柱,流动相为甲醇-水(0.5%冰醋酸)梯度洗脱,流速为0.4 mL•min-1,柱温为40℃,检测波长为365 nm。并利用HPLC-ESI-TOFMS对主要共有峰进行鉴定,采用电喷雾正离子源模式,N2流速7L•min-1,载气温度320 ℃,毛细管电压3 kV,轰击电压100 V,扫描质子范围m/z 100~1000。结果:不同产地狭叶柴胡地上部分有14个共有峰,各共有峰之间的分离度好;利用质谱信息与文献结合对共有峰中的10个色谱峰进行了结构确认。结论:利用建立的特征色谱可以对狭叶柴胡地上部分药材进行质量评价,为狭叶柴胡地上部分资源的综合利用开发提供了科学依据。

Objective:To establish the HPLC fingerprint of flavonoids of Bupleurum scorzoneri folium Willd aerial parts from different Habitats.Method:Kromasil C18 (4.6 mm×250 mm,5 µm)column was used. The mobile phase was composed of methanol -0.5% acetic acid with a nonlinear gradient elution.The flow rate was 0.4 mL.min-1 .UV detector was set at 365 nm and column temperature was 40℃. And most of common peaks were identified by HPLC-ESI- TOF MS,Electron spray ionization and positive ion mode was used,the flow and temperature of carrier gas(N2) were 7 L•min-1 and 320℃,respectively;the capillary voltage was 3000 V,the bombardment voltage was 100V,the spectra were recorded in the range of m/z 100-1000.Result: Bupleurum scorzoneri folium Willd aerial part contains 14 common peaks and every common peaks were completely separated. Ten components are identified by the analysis of the MS spectra as well as that reported in literatures. Conclusion:The fingerprint can be used to distinguish Bupleurum scorzoneri folium Willd aerial parts medicinal materials. This study will provide a scientific basis for the development of comprehensive utilization of Bupleurum scorzoneri folium Willd aerial parts.
