摘要 目的:比较思茅蛇菰、穗花蛇菰、疏花蛇菰、粗穗蛇菰、多蕊蛇菰5种蛇菰的化学成分,为今后此5种蛇菰的进一步研究和质量控制提供参考。方法:采用TLC、HPLC-DAD和LC-MS/MS方法联合对5种蛇菰的脂溶性成分和水溶性成分进行分析。TLC展开剂为氯仿-石油醚(60~90 ℃)(3∶2);HPLC采用Agilent Zorbax SB-C18(4.6 mm×150 mm,5 μm)色谱柱,以甲醇(A)-0.2%甲酸水溶液(B)为流动相,梯度洗脱[0~10 min,A-B(5∶95)→(10∶90);10~40 min,A-B(10∶90)→(20∶80);40~60 min,A-B(20∶80)→(30∶70);60~75 min,A-B(30∶70);75~100 min,A-B(30∶70)→(40∶60)],流速1 mL·min-1,检测波长306 nm;质谱检测参数:电喷雾离子源,负离子扫描模式,干燥气流速10 L·min-1,毛细管电压3.5 kV。结果:5种蛇菰样品的脂溶性成分和水溶性成分十分相似,共有的酚性成分含量有所不同。结论:本文研究结果有利于蛇菰类药材的合理使用和质量控制。

Abstract Objective:To study the chemical components of 5 species of genus Balanophora,including B. simaoensis,B. spicata,B. laxiflora,B.dioica and B.polyandra,for evaluating and quality control of these herbs.Methods:Liposoluble constituents and water-soluble constituents in 5 species of genus Balanophora were investigated by TLC,HPLC-DAD and HPLC-MS/MS methods.The TLC developer was made of chloroform-petroleum ether(60-90 ℃)(3∶2).The reversed phase HPLC system consisting of an Agilent Zorbax SB-C18(4.6 mm×150 mm,5 μm)and a mixture of methanol(A)-0.2% formic acid(B)with gradient elution as the mobile phase was adopted;The gradient elution program was 0-10 min,A-B(5∶95)→(10∶90);10-40 min,A-B(10∶90)→(20∶80);40-60 min,A-B(20∶80)→(30∶70);60-75 min,A-B(30∶70);75-100 min,A-B(30∶70)→(40∶60);The flow rate was 1 mL·min-1,and the detection wavelength was 306 nm.The MS detection parameters were as follow:Electrospray ion source with negative scan mode was adopted,cone gas flow rate was 10 L·min-1,the capillary voltage was 3.5 kV.Results:Liposoluble and water-soluble constituents in 5 species of genus Balanophora were very similar,but the contents of common phenolic components were different.Conclusion:The results in this paper is beneficial in quality control and rational use of genus Balanophora.
