青叶胆药材及饮片中獐牙菜苦苷和龙胆苦苷的含量测定___

摘要 目的:建立反相高效液相色谱法测定青叶胆药材及饮片中獐牙菜苦苷和龙胆苦苷的含量。方法:采用Diamonsil C18色谱柱(250 mm×4.6 mm,5 μm),以甲醇-水(23∶77)为流动相,流速l mL·min-1,检测波长242 nm。结果:獐牙菜苦苷和龙胆苦苷2种成分均达到基线分离,线性范围分别为30~300 μg·mL-1(r=0.9997)和77.9~779 μg·mL-1(r=0.9999);药材中2种成分的平均回收率为103.3%(RSD=1.0%)和97.1%(RSD=1.7%),饮片中2种成分的平均回收率为98.5%(RSD=1.9%)及97.6%(RSD=1.5%)。结论:采用RP-HPLC法同时测定青叶胆中獐牙菜苦苷和龙胆苦苷的含量,该方法简便、快速、准确,为2010年版中国药典一部增订青叶胆含量测定项做了研究。

Abstract  Objective:To establish a quantitative method for simultaneous determination of swertiamarin and gentiopicroside in Swertia mileensis and its cut crude drug by RP-HPLC.Method:The two constituents were base-isolated on the column of Diamonsil C18(250 mm×4.6 mm,5 μm),eluted with methanol-water(23∶77)at a flow rate of 1.0 mL·min-1.The detection wavelength was 242 nm.Results:Two constituents were base-isolated,and the linear ranges of swertiamarin and gentiopicroside were 30-300 μg·mL-1(r=0.9997)and 77.9-779 μg·mL-1(r=0.9999);The average recoveries(n=6)of the two components in Swertia mileensis were 103.3%(RSD=1.0%) and 97.1%(RSD=1.7%),and the average recoveries(n=6)of the two components in cut crude drug were 98.5%(RSD=1.9%)and 97.6%(RSD=1.5%),respectively.Conclusions:The application of RP-HPLC method can simultaneously determine the content of swertiamarin and gentiopicroside in Swertia mileensis.The method is convenient,rapid and reliable.It can offer references for the quality control of Swertia mileensis T. N. Ho et W. L. Shih.
