Abstract Objective:To establish an RP-HPLC method for the determination of sodium tanshinone ⅡA sulfonate in rabbits plasma.Method:Protein precipitation with acetonitrile was applied to purify plasma samples before RP-HPLC analysis.The content of sodium tanshinone ⅡA sulfonate in plasma samples was determined on Hypersil C18 column(250 mm×4.6 mm,10 μm)with KH2PO4(0.01 mol·L-1),with methyl alcohol and triethylamine(33∶67∶05,adjusted to pH 5.5± 0.1 with phosphoric acid)as the mobile phase,flow rate of 1 mL·min-1,detected at 271 nm and column temperature at room temperature.Result:The retention time of sodium tanshinone ⅡA sulfonate was observed at 6.6 min.The linear range of calibration curve was within drug concentrations of 0.1-30 mg·L-1(n=5,r=0.9994).The quantitation limit was 0.1 mg·L-1.The analytical recovery was more than 95%,intra-and inter-day precision RSDs were all less than 5.0%(n=5).The experimental data showed that the concentration-time curve of sodium tanshinone ⅡA sulfonate in rabbits plasma could be fitted to three-compartment model(W=1).Conclusion:These findings can provide references during pharmacokinetics investigations in humans for sodium tanshinone ⅡA sulfonate.
