Abstract Objective：To establish an HPLC method for the quantitative determination of saikosaponin b2 in Radix Bupleuri oral liquids.Method:Macroporous absorption resin was employed to purify saikosaponin b2.The HPLC method was performed on a Hypersil C18 column(250 mm×4.6 mm,5 μm)with gradient elution of acetonitrile-water(0-50 min,30∶70→50∶50),the flow rate was 1.0 mL·min-1,the column temperature was set at 30 ℃,and the detective wavelength was 252 nm.Result:The calibration curve was linear in the range of 0.03-0.30 μg(r=0.9998) for saikosaponin b2.The average recovery(n=6)was 98.0%(RSD=0.9%).Conclusion:This method is sensitive,simple and accurate,and can be used to determine the content of saikosaponin b2 in Radix Bupleuri oral liquids.
