《中华人民共和国药典》收载的鹿源药材中胶原蛋白对比分析

目的: 研究《中华人民共和国药典》收载的鹿源药材中胶原蛋白含量,以期为鹿源药材的质量评价和控制提供理论依据。方法: 参考GB 5009.5-2016方法,通过杜马斯燃烧法测定鹿源药材中蛋白质含量,折算系数为6.25;在NY/T 1618-2008方法基础上做略微改动,采用氨基酸自动分析法测定鹿源药材中4个构成胶原蛋白的氨基酸含量,进而折算胶原蛋白含量,折算系数为10。结果: 梅花鹿的鹿茸、鹿角、鹿角脱盘、鹿角胶、鹿角脱盘胶、鹿角霜和鹿角脱盘霜中的胶原蛋白含量分别为391.0、157.0、159.8、716.4、735.0、12.2和13.2 mg·g-1;马鹿的鹿茸、鹿角、鹿角脱盘、鹿角胶、鹿角脱盘胶、鹿角霜和鹿角脱盘霜中的胶原蛋白含量分别为382.4、154.6、157.1、708.8、714.3、10.9和12.8 mg·g-1。结论: 鹿源药材中胶原蛋白含量以鹿角胶最高,鹿茸其次,鹿角次之,鹿角霜最低。鹿源药材中4个构成胶原蛋白氨基酸和胶原蛋白含量差异很大,可以考虑以羟脯氨酸、脯氨酸、甘氨酸、丙氨酸和胶原蛋白含量作为鹿源药材的质量评价标准。

Objective: To study the contents of collagen in deer related medicines in Chinese Pharmacopoeia and to provide a theoretical basis for the quality evaluation and control of deer related medicines. Methods: Referring to the method of GB 5009.5-2016,the protein content of deer related medicines was determined by Dumas combustion method,and the conversion coefficient was 6.25. Referring to the method of NY/T 1618-2008 with minor changes,the four constituent collagen amino acids content of deer related medicines was determined by amino acid automatic analyzer,and then converted into collagen content. The conversion coefficient was 10. Results: The contents of collagen in Cervi Cornu Pantotrichum,Cervi Cornu,antlers plate,Cervi Cornus Colla,antlers plate colla,Cervi Cornu Degelatinatum and antlers plate degelatinatum of Cervus nippon Temminck were 391.0,157.0,159.8,716.4,735.0,12.2 and 13.2 mg·g-1,respectively. The contents of collagen in Cervi Cornu Pantotrichum,Cervi Cornu,antlers plate,Cervi Cornus Colla,antlers plate colla,Cervi Cornu Degelatinatum and antlers plate degelatinatum of Cervus elaphus Linnaeus were 382.4,154.6,157.1,708.8,714.3,10.9 and 12.8 mg·g-1,respectively. Conclusion: The content of collagen in Cervi Cornus Colla is the highest in deer related medicines,Cervi Cornu Pantotrichum is the second,Cervi Cornu is the third,and Cervi Cornu Degelatinatum is the lowest. The contents of four kinds of amino acids constituting collagen and collagen in deer related medicines were different. Hydroxyproline,proline,glycine,alanine and collagen contents can be considered as the quality evaluation criteria for deer related medicines.
