HPLC-DAD法同时测定骨松宝颗粒中5个成分

目的:建立HPLC法同时测定骨松宝颗粒中的5个成分。方法:采用岛津InertSustain C18色谱柱(250 mm×4.6 mm,5 μm),流动相为乙腈-0.1%磷酸水溶液,梯度洗脱;体积流量为1.0 mL·min-1;柱温30℃。结果:芒果苷、芍药苷、阿魏酸、淫羊藿苷、川续断皂苷Ⅵ质量浓度分别在0.512 5~128.1 μg·mL-1r=1.000)、4.842~1 211 μg·mL-1r=1.000)、0.159 0~39.76 μg·mL-1r=1.000)、3.252~812.9 μg·mL-1r=1.000)和10.56~2 641 μg·mL-1r=1.000)范围内线性关系良好,平均加样回收率(n=6)分别为96.8%(RSD=2.6%)、96.4%(RSD=0.67%)、94.5%(RSD=1.8%)、102.4%(RSD=0.90%)和96.0%(RSD=1.6%)。3批样品中芒果苷、芍药苷、阿魏酸、淫羊藿苷、川续断皂苷Ⅵ的含量分别为0.057 6~0.115 1、0.574 8~0.993 1、0.018 9~0.029 3、0.456 9~1.992 3、1.334 7-3.011 8 mg·g-1结论:建立的HPLC方法重复性好,操作简便,可应用于骨松宝颗粒的质量控制。

Objective:To establish an HPLC method for the simultaneous determination of five components in Gusongbao granules. Methods:The analysis was performed on a thermostatic Shimadzu InertSustain C18 column(250 mm×4.6 mm,5 μm)at 30℃,with the mobile phase comprising of acetonitrile-0.1% phosphoric acid at a flow rate of 1.0 mL·min-1 in a gradient elution manner. Results:The linear ranges of mangiferin,paeoniflorin, ferulic acid,icariin and asperosaponinⅥ were 0.512 5-128.1 μg·mL-1(r=1.000),4.842-1 211 μg·mL-1(r=1.000),0.159 0-39.76 μg·mL-1(r=1.000),3.252-812.9 μg·mL-1(r=1.000)and 10.56-2 641 μg·mL-1,respectively.(r=1.000). The average recoveries were 96.8%(RSD=2.6%),96.4%(RSD=0.67%),94.5%(RSD=1.8%),102.4%(RSD=0.90%)and 96.0%(RSD=1.6%),respectively. The contents of mangiferin,paeoniflorin,ferulic acid,icariin and asperosaponin Ⅵ in three batches of samples were 0.057 6-0.115 1,0.574 8-0.993 1,0.018 9-0.029 3,0.456 9-1.992 3 and 1.334 7-3.011 8 mg·g-1,respectively. Conclusion:The method is simple and repeatable,which can be used for the quality control of Gusongbao granules.
