凝胶剂中1株洋葱伯克霍尔德菌的分离、鉴定与生长特性研究

目的:对1株分离于凝胶剂的洋葱伯克霍尔德菌(Burkholderia cepacia)进行鉴定和生长特性研究。方法:通过生理生化和分子生物学方法进行鉴定。将鉴定为洋葱伯克霍尔德菌的菌株接种到《中华人民共和国药典》2015年版微生物限度检查法的培养体系中进行生长特性研究,并测定苯扎溴铵等对该菌的最小抑菌浓度和该菌的最低生长水分活度值。结果:该菌在药品检验用非选择性培养基中均能够生长良好,其最适生长培养基为寡营养培养基R2A培养基;在选择性培养基麦康凯液体、麦康凯琼脂和紫红胆盐琼脂培养基中能够生长良好。苯扎溴铵对该菌的最小抑菌浓度(MIC)为64 μg·mL-1,该菌最低生长水分活度为0.95~0.98。结论:该凝胶剂中污染的洋葱伯克霍尔德菌有进一步繁殖的可能,提示该产品存在一定的安全隐患,应进行风险评估并采取措施。

Objective:To identify and investigate the growth characteristics of a Burkholderia cepacia in gel. Methods:Bacteria were identified by using physiological biochemical methods and molecular biological methods. The strain identified as Burkholderia cepacia was inoculated into the cuture system in according to the ChP 2015 microbial limit test for its growth characteristics. Meanwhile,the minimum inhibitory concentration of benzalkonium bromide and the minimum level of growth water activity for this bacterium were determined. Results:This bacterium was able to grow well in non-selective media for drug testing,and the optimal growth medium was oligotrophic R2A medium;selective medium,for instance,this bacterium could grow well in MacConkey broth,MacConkey agar and violet red bile glucose agar. The minimum inhibitory concentration(MIC)of benzalkonium bromide to this bacterium was 64 μg·mL-1,and the minimum growth water activity for it was 0.95-0.98. Conclusion:The contaminated Burkholderia cepacia may have further bred in the gel,suggesting this product potentially has certain safety hazard as risk assessment and measures should be carried out.
