非靶标代谢组学研究的影响因素分析

代谢组学技术是近10年来国际上提出和发展的新型组学技术,是系统生物学的重要组成部分。其中,非靶标代谢组学是研究最早且应用最广的方法。然而,由于代谢物种类多,理化性质差异大,加之样本对环境变化较敏感,导致了样本从收集、处理到储存过程的波动都会对代谢轮廓的分析产生一定影响。此外,代谢组学研究还涉及分析科学以及化学计量学等专业知识的应用。如何应用现代高通量分析技术(核磁共振和质谱)高精度快速对大量样本进行分析,对海量的原始数据进行预处理及挖掘,对差异代谢物准确定性和定量分析,是非靶标代谢组学研究现存的难点和热点。本文综述分析了近年来非靶标代谢组学研究过程中的一些影响因素及应对策略。

Metabolomics is a new developed omics technology in the world in recent ten years,and is an important part of system biology.Among them,untargeted metabolomics is the earliest and most widely used method.However,due to the variety of metabolites,the difference of physical and chemical properties,and the collection,processing and storage of samples have a certain effect on the analysis of metabolic profile due to their sensitive to the environment.In addition,metabolomics also involves the application of analytical science and chemometrics.How to apply modern high-throughput analysis technology (NMR and MS)to analyze a large number of samples quickly and accurately,and preprocess and mine massive raw data,and accurately,qualitatively and quantitatively analyze different metabolites are existing difficulties and hotspots in untargeted metabolomics studies.This paper reviewed and analyzed some influencing factors and strategies in the research process of untargeted metabolomics in recent years.
