星点设计-响应面法优化枳实中黄酮类化合物的水热法提取工艺

目的:利用星点设计-响应面法优化枳实中黄酮类化合物的水热法提取工艺。方法:以黄酮类化合物(柚皮苷、橙皮苷和新橙皮苷)的得率为评价指标,通过单因素试验,研究提取温度、液料比及提取时间对枳实中黄酮类化合物得率的影响,采用响应面法优选水热法的最佳提取条件。结果:水热法提取黄酮类化合物的最佳条件:提取温度114℃,液料比66∶1,提取时间50 min;在此条件下黄酮类化合物的得率为19.82%,与预测值的偏差为0.56%。结论:采用星点设计-响应面法优选的水热法提取工艺,方法简便,预测性良好,为黄酮类及中药活性物质的提取提供了一条新途径。

Objective:To optimize the extracting process for the flavonoids from Fructus Aurantii Immaturus by central composite design and response surface method.Methods:The single factor experiment combined with central composite design was used to optimize the hydrothermal extraction technology,with the yield of flavonoids (naringin,hesperidin and neohesperidin)as evaluation index.The three factors,including the extraction temperature,liquid/solid ratio and extraction time,were optimized for the hydrothermal extraction process of flavonoids from Fructus Aurantii Immaturus.Results:The results showed that the optimum hydrothermal extraction parameters:extraction temperature 114℃,liquid/solid ratio 66:1,extraction time 50 min.Under the conditions,the yield of flavonoids was 19.82%.The deviation between observed and predicted value was 0.56%.Conclusion:The central composite design and response surface method is convenient and highly predictive for optimizing the hydrothermal extraction process of the flavonoids from Fructus Aurantii Immaturus. This extraction method is not only a new technology of flavonoids extraction,but also provides a new idea to extract the active substances from tranditional Chinese medicine.
