超高效液相色谱法同时测定三七的叶和花中3个皂苷类成分含量

目的:建立超高效液相色谱法测定三七的叶和花中人参皂苷Rb1、Rb2和Rb3的含量。方法:采用Waters ACQUITY BEH C18色谱柱(2.1 mm×100 mm,1.7 μm),以乙腈-水为流动相进行梯度洗脱,流速0.4 mL·min-1,柱温30℃,检测波长203 nm。结果:人参皂苷Rb1、Rb2和Rb3线性关系良好;三七叶中上述3个成分平均加样回收率(n=9)均为100.5%,三七花中上述3个成分平均加样回收率(n=9)分别为99.3%、99.7%、98.6%。12批三七叶样品中人参皂苷Rb1、Rb2和Rb3含量范围分别为0.35%~0.59%、0.16%~0.68%、0.73%~3.33%,11批三七花样品中上述3个成分含量范围分别为0.46%~2.91%、0.83%~2.36%、2.95%~8.37%。结论:建立的方法可用于三七叶和三七花的含量测定。

Objective:To establish a method for determination of ginsenoside Rb1,ginsenoside Rb2 and ginsenoside Rb3 in leaves and flower buds of Panax notoginseng by UPLC. Methods:The analysis was performed on a Waters ACQUITY BEH C18 column(2.1 mm×100 mm,1.7 μm),with acetonitrile(A)-water(B) as the mobile phase with gradient elution at a flow rate of 0.4 mL·min-1. The column temperature was 30and the detection wavelength was 203 nm. Results:The linear relationships of ginsenoside Rb1,ginsenoside Rb2 and ginsenoside Rb3 were favorable;the recoveries(n=9) of the above-mentioned components in leaves of Panax notoginseng were 100.5%,100.5% and 100.5%,respectively and the recoveries(n=9) of the above-mentioned components in flower buds of Panax notoginseng were 99.3%,99.7% and 98.6%%,respectively. The contents of ginsenoside Rb1,ginsenoside Rb2 and ginsenoside Rb3 in 12 batches of leaves of Panax notoginseng were 0.35%-0.59%,0.16%-0.68% and 0.73%-3.33%,respectively. The contents of the three components in 11 batches of flower buds of Panax notoginseng samples were 0.46%-2.91%,0.83%-2.36% and 2.95%-8.37%,respectively. Conclusion:The established method can be used for determination of leaves and flower buds of Panax notoginseng.
