目的:建立同时测定麻杏止咳糖浆中盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷、甘草苷、甘草酸含量的高效液相色谱方法。方法:采用Thermo C18色谱柱(250 mm×4.6 mm,5 μm),以乙腈-0.02 mol·L-1磷酸二氢钾溶液(含0.1%三乙胺和0.1%磷酸)为流动相,梯度洗脱,流速1.0 mL·min-1,柱温25℃,检测波长分别为210 nm(麻黄碱、伪麻黄碱、苦杏仁苷)、237 nm(甘草苷、甘草酸)。结果:盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷、甘草苷、甘草酸进样量分别在9.994×10-2~9.994 μg(r=0.999 2)、4.388×10-2~4.388 μg(r=0.999 0)、0.119 9~11.99 μg(r=0.999 7)、6.794×10-2~6.794 μg(r=0.999 5)、4.308×10-2~4.308 μg(r=0.999 3)的范围内与峰面积积分值呈良好的线性关系;平均回收率分别为98.8%、101.4%、98.0%、98.3%、102.0%,RSD分别为1.2%、1.5%、1.7%、2.0%、0.99%。3批麻杏止咳糖浆样品中盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷、甘草苷、甘草酸的含量测定结果分别为0.471~0.501、0.194~0.219、0.523~0.542、0.141~0.150、0.307~0.330 mg·mL-1。结论:所建方法可用于测定麻杏止咳糖浆中盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷、甘草苷、甘草酸的含量。
Objective:To establish an HPLC method for simultaneous determination of five components (ephedrine hydrochloride,pseudoephedrine hydrochloride,amygdalin,glycyrrhizin and glycyrrhizic acid)in Maxing Zhike syrup.Methods:The analysis was performed on a Thermo C18 column (250 mm×4.6 mm,5 μm),using acetonitrile-0.02 mol·L-1 of potassium dihydrogen phosphate solution (containing 0.1% triethylamine and 0.1% phosphoric acid)as mobile phase under gradient elution and the flow rate was 1.0 mL·min-1.The column temperature was 25℃ and the detective wavelength was 210 nm for ephedrine,pseudoephedrine and amygdalin and 237 nm for glycyrrhizin and glycyrrhizic acid.Results:The linear ranges of ephedrine hydrochloride,pseudoephedrine hydrochloride,amygdalin,glycyrrhizin and glycyrrhizic acid were 9.994×10-2-9.994 μg (r=0.999 2),4.388×10-2-4.388 μg (r=0.999 0),0.119 9-11.99 μg (r=0.999 7),6.794×10-2-6.794 μg (r=0.999 5)and 4.308×10-2-4.308 μg (r=0.999 3),respectively;the average recoveries were 98.8%,101.4%,98.0%,98.3% and 102.0% with RSDs of 1.2%,1.5%,1.7%,2.0% and 0.99%,respectively.The contents of above-mentioned compounds in 3 samples were 0.471-0.501 mg·mL-1 for ephedrine hydrochloride,0.194-0.219 mg·mL-1 for pseudo ephedrine hydrochloride,0.523-0.542 mg·mL-1 for amygdalin,0.141-0.150 mg·mL-1 for glycyrrhizin and 0.307-0.330 mg·mL-1 for glycyrrhizic acid.Conclusion:The established method can be used for determination of ephedrine hydrochloride,pseudoephedrine hydrochloride,amygdalin,glycyrrhizin and glycyrrhizic acid in Maxing Zhike syrup.
