银杏叶总黄酮醇苷含量快速测定方法研究

目的:建立一种供试品处理简单的银杏叶总黄酮醇苷含量测定方法。方法:采用单因素试验,考察溶剂量、提取时间、提取温度的影响,以总黄酮醇苷含量为评价指标,筛选最佳提取方法,并对该方法进行方法学考察及验证。结果:溶剂量80 mL,提取时间45 min,回流温度90℃为最佳实验条件;槲皮素、山柰酚、异鼠李素分别在0.158~15.78 μg、0.109~10.87 μg、0.019~1.944 μg范围内呈良好的线性关系(r≥0.999 0);槲皮素、山柰酚、异鼠李素的平均加样回收率分别为95.4%、93.8%、92.0%,符合限度要求,该方法加样回收率良好(RSD均<3%);15批银杏叶验证结果与《中华人民共和国药典》(以下简称中国药典)2015年版方法测定结果不具有显著差异[相对平均偏差(relative average deviation,RAD)均<3%]。结论:本试验建立的方法准确可行,操作简便,较中国药典方法效率高,有助于实时监测银杏叶质量,且该方法节省时间和溶剂,减少了资源浪费和环境污染,同时也为2020年版中国药典修订提供参考依据。

Objective:To establish a content determination method of total flavonoid glycosides in Ginkgo biloba,with simple preparation.Methods:Taking total flavonoid glycosides as an assessment index,the best extractive method (including solvent volume,extraction time and extraction temperature) was selected with single factor experiments.And the methodology was verified.Results:The solvent volume of 80 milliliters,extracting time in 45 minutes and reflow temperature at 90℃,showed the best experimental conditions.The method showed good linearity in the range of 0.158-15.78μg for quercetin,0.109-10.87μg for kaempferol and 0.019-1.944μg for isorhamnetin (r ≥ 0.999 0).The average recoveries were 95.4%,93.8% and 92.0% for quercetin,kaempferol and isorhamnetin respectively with RSDs<3%,all of which met the limit requirements of ChP.Making a simultaneous determination of the fast determination method and ChP method for 15 batches of ginkgo biloba,the results showed no significant difference (relative average deviation <3%).Conclusion:The established method is accurate,feasible and easy,which is more efficient than pharmacopoeia method.It helps to monitor the quality of Ginkgo biloba constantly.Besides,the method is time and solvent saving,which can reduce waste and environmental pollution.Meanwhile,it provides a reference for revision of the 2020 edition ChP.
