目的:采用HPLC法同时测定锥叶柴胡中6个皂苷类成分柴胡皂苷A、C、D及柴胡次皂苷F、G、I的含量。方法:采用InertSustain AQ-C18色谱柱(4.6 mm×250 mm,5 μm),以乙腈-水为流动相进行梯度洗脱,流速1.0 mL·min-1,检测波长210 nm,柱温25℃。结果:柴胡皂苷A、C、D及柴胡次皂苷F、G、I质量浓度分别在8.32~208、3.50~88.2、24.0~600、13.2~331、31.3~783、6.65~166 μg·mL-1范围内呈良好的线性关系(r≥0.999);平均回收率为91.1%~107.7%(RSD<3%,n=6)。12批样品中上述6个成分含量分别为0.11%~0.34%、0.05%~0.20%、0.35%~1.92%、0.13%~0.87%、0.18%~1.49%、0.04%~0.37%。结论:新建立的含量测定方法准确可靠,可用于锥叶柴胡的质量控制。
Objective: To establish an HPLC method for simultaneous determination of saikosaponin A,saikosaponin C,saikosaponin D,prosaikogenin F,prosaikogenin G and prosaikogenin I in Bupleurum bicaule Helm.Methods: HPLC analysis was performed on an InertSustain AQ-C18 column (4.6 mm×250 mm,5 μm) with a gradient elution of acetonitrile and water.The flow rate was 1.0 mL·min-1,the detection wavelength was 210 nm and the column temperature was 25℃.Results: The linear ranges of saikosaponin A,saikosaponin C,saikosaponin D,prosaikogenin F,prosaikogenin G and prosaikogenin I were 8.32-208 μg·mL-1,3.50-88.2 μg·mL-1,24.0-600 μg·mL-1,13.2-331 μg·mL-1,31.3-783 μg·mL-1,6.65-166 μg·mL-1,respectively.The average recoveries were 91.1%-107.7%(RSD<3%,n=6).The contents of the six components in 12 samples of B.bicaule were 0.11%-0.34%,0.05%-0.20%,0.35%-1.92%,0.13%-0.87%,0.18%-1.49%,0.04%-0.37%,respectively.Conclusion: The newly established method is accurate and reliable,which can be used for the quality control of B.bicaule.
