目的:建立RP-HPLC法测定黄连上清胶囊中6个生物碱的含量。方法:色谱条件:采用Agilent Eclipse XDB C18色谱柱(250 mm&#215;4.6 mm,5 μm),乙腈-50 mmol&#183;L-1磷酸二氢钾水溶液(40:60)(混合溶液中加15 mmol&#183;L-1十二烷基磺酸钠,再以磷酸调节pH至4.0)为流动相;流速1.0 mL&#183;min-1;检测波长345 nm;柱温35℃。结果:在该色谱条件下,6个生物碱的分离度良好;在指定浓度范围内线性关系均良好;精密度与重复性RSD均<2.0%;样品溶液配制后24 h内稳定性较好;平均回收率均大于98%,RSD均<2.0%。3批黄连上清胶囊中6个生物碱总量分别为1 140、1 204、1 461 μg&#183;粒-1结论:该方法准确、简便、可靠,分离效果好,可用于黄连上清胶囊的质量评价,为黄连上清胶囊的质量标准的提高提供依据。

Objective: To determine six alkaloids in Huanglian Shangqing capsules by RP-HPLC.Method: The chromatographic analysis were carried out on an Agilent Eclipse XDB C18 column (250 mm&#215;4.6 mm,5 μm).The mobile phase was acetonitrile-50 mmol&#183;L-1 monopotassium phosphate(40:60)(containing 15 mmol&#183;L-1 SDS and adjusting to pH=4.0 with phosphoric acid) at the flow rate of 1.0 mL&#183;min-1.The detective wavelength was set up at 345 nm and the column temperature was 35℃.Results: Under these chromatographic conditions, six alkaloids were well separated;the linear relationship was good within the specified range;the precision and repeatability RSD was less than 2.0%;the stability of the sample solution was good within 24 hours after preparation;the average recovery was greater than 98%, RSD was<2.0%.The total amount of 6 alkaloids in the three batches of Huanglian Shangqing capsules were 1 140 μg&#183;capsule-1,1 204 μg per capsule and 1461 μg per capsule,respectively.Conclusion: The method is accurate, simple, reliable and effective, and can be used for the quality evaluation of Huanglian Shangqing capsules, which provides the basis for improving the quality standard of Huanglian Shangqing capsules.
