UFLC-MS/MS法分析不同来源白芷中9个新型呋喃香豆素

目的:超高速液相色谱-质谱联用(UFLC-MS/MS)技术分析不同来源白芷中9个新型呋喃香豆素(杭白芷香豆素A、B、C、D、E、F、G、H、J)的含量。方法:甲醇超声提取白芷,提取物经UFLC-MS/MS分析,色谱柱为Kinetex® C18(100 mm×2.1 mm,2.6 μm),流动相为乙腈和水,梯度洗脱,流速为0.5 mL·min-1。选择电喷雾离子源,在正离子模式下进行检测,定量分析选用多反应监测模式。对来源于重庆市南川县、安徽省亳州市、湖南省郡东县、吉林省通化市、黑龙江省绥棱县、中国台湾台中市和花莲县样品中的上述9个成分的含量进行测定。结果:经过优化,9个呋喃香豆素可在12 min内被准确定量。系统适用性、线性、检测下限、定量下限、精密度、回收率、稳定性经验证良好。不同产地白芷中上述9个成分含量变化差异极大。结论:该方法准确、高效,灵敏度高,可用于白芷中9个新型呋喃香豆素的含量测定。

Objective:To determine the contents of nine furanocoumarins (andafocoumarins A,B,C,D,E,F,G,H,J) in Angelicae Dahuricae Radix (ADR) from different regions using ultra-fast liquid chromatography coupled with triple quadrupole mass spectrometer (UFLC-MS/MS).Methods:UFLC-MS/MS was used to analyze ADR extracts obtained by ultrasonic treatment with methanol.A Kinetex® C18 column (100 mm×2.1 mm,2.6 μm) was used to carried out the separation with a gradient elution by acetonitrile and water.The flow rate was 0.5 mL·min-1.All the samples were determined using positive electrospray ionization with multiple reaction monitor mode.The contents of above-mentioned 9 ingredients in samples originated from Nanchuan county of Chongqing city,Bozhou city of Anhui province,Jundong county of Hunan province,Tonghua city of Jilin province,Suileng county of Heilongjiang province,as well as Taichung city and Hualien county of Taiwan province in China were determined.Results:Through a series of optimization,all the above-mentioned 9 analytes could be accurately quantified within 12 min.System suitability,linearity,lower limit of detection,lower limit of quantification,precision,recovery and stability of the method were verified.Some of these 9 andafocoumarins were significantly different in different regions of ADR.Conclusion:The method is accurate,efficient and sensitive and can be used to determine the contents of 9 furanocoumarins in ADR.
