非水反相色谱-飞行时间质谱法对鱼肝油中维生素A酯的分离与鉴定

目的:建立非水反相色谱-飞行时间质谱法对鱼肝油中维生素A酯组分进行分离鉴定。方法:选择C18色谱柱(250 mm×4.6 mm,5 μm),以乙腈-异丙醇(8:2)为流动相,对鱼肝油中的维生素A酯组分进行分离,并结合对照品比对与电喷雾正离子化/飞行时间质谱法进行结构鉴定。结果:在本试验研究的3种样品中,不同鳕鱼肝油样品可分别检测到维生素A醋酸酯与维生素A棕榈酸酯。金枪鱼肝油样品中共鉴定了15种维生素A酯,以维生素A油酸酯、棕榈酸酯、棕榈油酸酯、二十碳五烯酸(EPA)酯与二十二碳六烯酸(DHA)酯相对含量较高。结论:本文应用的方法能有效地分离鉴定鱼肝油中维生素A酯组分,可为鱼肝油质量检测提供参考。

Objective:To develop a non-aqueous reverse phase high performance liquid chromatography (NARP)-TOF/MS method for the separation and identification of retinyl esters in fish liver oil.Methods:The retinyl esters were separated on a C18(250 mm×4.6 mm,5 μm) column with acetonitrile-isopropanol (8:2) as the mobile phase.The structures were elucidated by comparing with reference substances and by ESI+-TOF/MS.Results:Retinyl acetate and retinyl palmitate were found in cod liver oil. 15 kinds of retinyl esters were found in tuna liver oil,with retinyl oleate,retinyl palmitate,retinyl palmitoleate,eicosapentenoic acid (EPA) retinyl ester and docosahexenoic acid (DHA) claiming the higher contents.Conclusion:The established method is useful for the separation and identification of retinyl esters in fish liver oil,thus providing reference for quality control of the product.
