HPLC法测定藏药宽筋藤中3个生物碱类成分的含量

Content determination of three alkaloid constituents in Tibetan medicine Tinosporae Sinenisis Caulis by HPLC

目的:建立高效液相色谱法同时测定宽筋藤药材中药根碱、巴马汀和小檗碱3个生物碱成分的含量,并对14个不同地区商品药材进行含量测定。方法:采用Sunfire C18色谱柱(4.6 mm×250 mm,5μm),柱温30℃,以乙腈(A)-水(0.05 mol·L-1磷酸二氢钾溶液,加0.1%磷酸) (B)为流动相梯度洗脱(0~15 min,20%A→32%A;15~25 min,32%A),流速1.2 mL·min-1,检测波长340 nm。结果:盐酸药根碱、盐酸巴马汀和盐酸小檗碱3种成分质量浓度分别在0.17~0.86、0.08~0.40和0.05~0.26 μg·mL-1范围内呈良好线性关系(r ≥ 0.9997),平均回收率(n=6)分别为99.3%、101.6%和100.0%。14个不同地区16批商品药材含量测定结果显示3个成分含量差异较大。结论:所建立的方法经方法学验证,可用于宽筋藤药材的质量控制和评价。

Objective: To establish an HPLC method for simultaneously determining the content of three alkaloids including jatrorrhizine, palmatine and berberine in Tinosporae Sinenisis Caulis Herba and the contents of the commercial medicinal materials of Tinosporae Sinenisis Caulis collected from fourteen different producing areas have been determined.Methods:HPLC was performed on a Sunfire C18 column (4.6 mm×250 mm, 5 μm) at 30℃ with acetonitrile(A)-water (0.05 mol·L-1 potassium dihydrogen phosphate, 0.1% phosphoric acid added) (B) as the mobile phase by gradient elution (0-15 min, 20%A→32%A; 15-25 min, 32%A) at a flow rate of 1.2 mL·min-1, and the detection wavelength was at 340 nm.Results:Good linear relationship was shown in the range of 0.17-0.86, 0.08-0.40 and 0.052-0.260 μL·mL-1 of jatrorrhizine hydrochloride, palmatine hydrochloride and berberine hydrochloride, respectively (r≥0.999 7),and the average recoveries (n=6) were 99.3%, 101.6% and 100.0%, respectively. Results of the content determination for commercial medicinal materials of total sixteen batches from fourteen different areas showed that there were significant differences in the contents of the three constituents.Conclusion:The established method could provide a reliable approach to the control and evaluation of the quality of Tinosporae Sinenisis Caulis.
