目的:建立同时测定旱柳叶药材中木犀草苷、芹菜素-7-O-β-D-吡喃葡萄糖醛酸苷和木犀草素-3’-甲醚-7-O-β-D-吡喃葡萄糖醛酸苷含量的高效液相色谱法。方法:采用YMC-Pack ODS-A(150 mm×4.6 mm,5 μm)色谱柱,以乙腈-0.2%磷酸溶液(15:85)为流动相,流速1.0 mL·min-1,检测波长344 nm。结果:木犀草苷,芹菜素-7-O-β-D-吡喃葡萄糖醛酸苷和木犀草素-3’-甲醚-7-O-β-D-吡喃葡萄糖醛酸苷进样量分别在0.26~2.08 μg(r=0.9999)、0.30~2.40 μg(r=0.9999)和0.62~4.96 μg(r=0.9999)范围内与峰面积呈良好线性关系;平均回收率(n=6)分别为97.5%、98.8%和101.5%,RSD分别为1.7%、1.7%和1.6%。结论:经方法学验证,本法可用于旱柳叶药材的质量控制。

Objective: To establish a method for the simultaneous determination of the contents of luteolin-7-O-β-D-glucoside,apigenin-7-O-β-D-pyranglycuronide and lutelion-3'-methylether-7-O-β-D-pyranglycuronide in Hankow Willow Leaf by HPLC. Methods: The separation was performed on a YMC-Pack ODS-A column (150 mm×4.6 mm,5 μm) with a mobile phase of acetonitrile-0.2% phosphoric acid solution (15:85).The flow rate was 1.0 mL·min-1,and the detecting wavelength was 344 nm. Results: The linear ranges of luteolin-7-O-β-D-glucoside,apigenin-7-O-β-D-pyranglycuronide and lutelion-3'-methylether-7-O -β-D-pyranglycuronide were 0.26-2.08 μg (r=0.9999),0.30-2.40 μg (r=0.9999) and 0.62-4.96 μg (r=0.9999),respectively.The average recoveries (n=6) were 97.5% (RSD=1.7%),98.8% (RSD=1.7%) and 101.5% (RSD=1.6%),respectively. Conclusion: Based on method validation,the method can be used for quality control of the leaves of Salix matsudana Koidz.
