RP-HPLC法测定郁李仁中郁李仁苷A和阿福豆苷含量___

目的: 建立同时测定不同产地、不同种属郁李仁中郁李仁苷A和阿福豆苷含量的RP-HPLC分析方法。 方法: 采用Kromasil C₁₈(250 mm×4.6 mm,5 μm)色谱柱,流动相为甲醇-0.1%醋酸水溶液(52:48,v/v),流速1.0 mL·min^-1,检测波长264 nm,柱温35 ℃。 结果: 郁李仁苷A和阿福豆苷浓度分别在2.512~40.19 μg·mL^-1(r=0.9997),0.996~15.94 μg·mL^-1(r=0.9995)范围内与峰面积呈现良好的线性关系;平均回收率(n=9)分别为98.8%和100.2%。 结论: 此方法快速、准确,重复性好,可作为测定不同种属郁李仁药材中郁李仁苷A和阿福豆苷含量的方法。

Objective: To establish an RP-HPLC method for the determination of prunuside A and afzelin in Semen Pruni of different species and different areas. Methods: The chromatographic method was carried out on Kromasil C₁₈ column(250 mm×4.6 mm,5 μm)using methanol-0.1% acetic acid solution(52:48,v/v)as the mobile phase;The flow rate was 1.0 mL·min^-1;the detection wavelength was set at 264 nm,and the column temperature was 35 ℃. Results: The linear range were 2.512-40.19 μg·mL^-1(r=0.9997)for prunuside A and 0.996-15.94 μg·mL^-1(r=0.9995)for afzelin respectively.The average recoveries(n=9)were 98.8% and 100.2%. Conclusion: The method is simple,accurate with good reproducibility.It can be applied in quantitative of determination of prunuside A and afzelin in Semen Pruni.
