RP-HPLC法测定郁李仁中苦杏仁苷含量___

摘要 目的：建立RP-HPLC法测定不同产地、不同种属郁李仁中苦杏仁苷含量的方法。方法：采用Kromasil C18柱（5 um，250 mm × 4.6 mm）；以乙腈﹣水（12 : 88, v/v）为流动相；柱温为35 ºC；检测波长210 nm；流速为1.0 mL • min-1。结果：苦杏仁苷的线性范围为5.040～80.6 ug • mL-1（r = 0.9999，n = 6）；平均回收率（n = 9）为99.8%，RSD为2.3%。结论：本法简便，结果准确，重复性好，可作为测定不同种属郁李仁药材中苦杏仁苷含量的方法。

Abstract Objective: To establish an RP-HPLC method for the determination of Amygdalin in different species of Semen Pruni in different areas.Methods: The chromatographic method was carried out on Kromasil C18 colume （5 um, 250 mm × 4.6 mm）using acetonitrile﹣water（12 : 88, v/v）as the mobile phase; the colume temperature at 35 ºC; the detection wavelength was set at 210 nm and the flow rate was 1.0 mL • min-1. Results: The linear range was 5.040～80.6 ug • mL-1（r = 0.9999, n = 6）for Amygdalin.The average recoveries (n = 9) was 99.8%.Conclusion: The method is simple,accurate with good reproducibility.It can be applied in quantitative of determination of Amygdalin in Semen Pruni.
