摘要 目的:研究更昔洛韦(GCV)在碳糊电极(CPE)上的电化学行为、电化学动力学性质、可能的电极反应机理和电化学定量分析方法。方法:运用循环伏安法(CV)、计时电流法(CA)、计时电量法(CC)和差分常规脉冲伏安法(DNPV)。结果:GCV在CPE上的伏安行为是一受扩散控制的不可逆电化学氧化过程,氧化峰电位(Ep)为1.191 V,并且扫描速度在5~1000 mV·s-1范围内Ip与v1/2呈良好的线性关系。测得GCV在CPE上的电极过程动力学参数:电子转移系数α 为0.44;扩散系数D为8.86×10-4 cm2·s-1;电极反应速率常数kf为2.10×10-2 s-1。用DNPV法测得GCV氧化峰电流与其浓度在5.0×10-7~4.0×10-4 mol·L-1范围内呈良好的线性关系(r=0.9986),检出限为4.2×10-8 mol·L-1,RSD为3.0%~4.0%,加样回收率为98.0%~100.4%。结论:本方法仪器简单,操作便捷,分析成本低廉,用于GCV含量测定结果令人满意。

Abstract Objective:Electrochemical behaviors,electrochemical kinetic parameters and its reaction mechanism of ganciclovir(GCV) at carbon paste electrode(CPE) were investigated. Methods:Cyclic voltammetry(CV),chronoamperometry(CA),chronocoulometry(CC) and differential normal pulse voltammetry(DNPV).Results:Electrochemical experimental results show that GCV electrochemical behavior in 0.10 mol·L-1 PBS exhibits an irreversible electrochemical redox process at CPE,the oxidation peak potential(Ep) is 1.191 V(vs.SCE).The plot of oxidation peak currents versus the square roots of the scanning rates for the redox in the potential range of 5-1000 mV·s-1 show a straight line, as expected for a diffusionlimited electrochemical process for GCV electrochemical oxidation.Electron transfer coefficient α,diffusion coefficient D and the apparent reaction rate constant kf are 0.44,8.86×10-4 cm2·s-1 and 2.10×10-2 s-1,respectively.GCV shows an irreversible process with one electron and one proton transferred at CPE.The changes of the oxidation peak currents with GCV concentration were examined by DNPV.The oxidation peak currents of GCV versus its concentration have a good linear relationship in the concentration range of 5.0×10-7 -4.0×10-4 mol·L-1 with the correlation coefficient of 0.9986 and the detection limit of 4.2×10-8 mol·L-1 by DNPV response.The relative standard deviation is between 3.0%-4.0% and recovery is in the range of 98.0%-100.4%.Conclusion:The proposed method can be applied in GCV electrochemical determination with low detection limit,high sensitivity and the satisfied results.
