目的:建立一测多评法同时测定西洋参类保健食品中9个活性成分(人参皂苷Rg1、Re、Rg2、Ro、Rb1、Rc、Rb2、Rb3、Rd)的含量。方法:采用Agilent Zorbax SB-Aq C18色谱柱(4.6 mm×250 mm,5 μm),0.005%甲酸水溶液(A)-0.005%甲酸乙腈溶液(B)为流动相,梯度洗脱(0~20 min,79% A;20~49 min,79% A→66% A;49~65 min,66% A→60% A),柱温30℃,流速为1.0 mL·min-1,检测波长203 nm。以人参皂苷Rb1为参照物,建立与其他8个成分的相对校正因子,计算其含量。并与外标法实测值比较差异。结果:在一定的线性范围内,人参皂苷Rb1与Rg1、Re、Rg2、Ro、Rc、Rb2、Rb3,Rd的RCF值分别为0.651 3、1.073、1.601、1.333、0.983 0、1.247、1.160、1.347,且在不同实验条件下重现性良好(RSD<5.0%)。外标法与一测多评法测定各成分含量无显著差异。结论:建立的一测多评法准确性高,可用于西洋参保健食品中皂苷类成分的定量分析及质量评价。
Objective: To establish a method for simultaneous determination of nine main active constituents(ginsenosides Rg1,Re,Rg2,Ro,Rb1,Rc,Rb2,Rb3 and Rd) in health foods containing American ginseng by quantitative analysis of multi-components by single marker(QAMS).Method: The chromatographic separation was achieved on an Agilent Zorbax SB-Aq C18 column(250 mm×4.6 mm,5 μm) and the mobile phase consisted of solvent A(0.005% formic acid water solution) and solvent B(0.005% formic acid acetonitrile solution) with gradient elution(0-20 min,79%A;20-49 min,79%A→66%A;49-65 min,66%A→60%A).The column temperature was 30.The flow rate was 1.0 mL·min-1.The UV detection wavelength was set at 203 nm. Ginsenoside Rb1 was used as the reference,the relative calibration factors of other 8 constituents to ginsenoside Rb1 were calculated respectively.And the contents of active constituents were calculated based on the relative calibration factors.The results were compared with those obtained by the external standard method. Results: Relative correction factors of ginsenoside Rg1,Re,Rg2,Ro,Rc,Rb2,Rb3 and Rd with reference to ginsenoside Rb1 were 0.651 3,1.073,1.601,1.333,0.983 0,1.247,1.160,and 1.347, respectively,and the repeatability was good in different experimental conditions with RSDs below 5%.There was no significant difference between the results determined by QAMS and the external standard method.Conclusion: The established QAMS method is accurate and can be used for quantitative analysis and quality evaluation of health foods containing American ginseng.
